Phagocytic activity of the leukemic cell and its response to the phagocytosis-stimulating tetrapeptide, tuftsin.

چکیده
tuftsin activity in the sera of patients was also evaluated. Fifteen patients were included in this study: six with myelofibrosis, seven with acute granulocytic leukemia, and two with myelomonocytic leukemia. In all six cases of myelofibrosis, the polymorphonuclear cells showed a normal level of basal phagocytosis in Krebs-Ringer phosphate glucose medium. Five of seven cases of acute granulocytic leukemia showed normal or near-normal values also, while two had significantly diminished levels. All thirteen of these patients failed to show stimulation with saturating amounts of autologous serum or tuftsin. This is distinctly abnormal, since normal blood granulocytes of humans, dogs, and rabbits are considerably stimulated. The two cases of myelomonocytic leukemia available for study showed a higher than normal basal phagocytic activity in Krebs-Ringer phosphate glucose medium and responded normally to serum and tuftsin stimulation.

The correlation between zinc and monocyte phagocytosis in patients with major b-thalassemia
Background: Zinc depletion decreases monocyte functions and survival while excessive amount of zinc inhibits monocyte activation. Monocytes shift from conducting intercellular communication to becoming innate immune function as a response. This study aims to examine the influence of zinc status on the monocyte phagocytosis in patients with major beta-thalassemia. Materials and Methods: This st...
متن کاملThe effect of Tuftsin on the nitrous blue tetrazolium reduction of normal human polymorphonuclear leukocytes.
The influence of Tuftsin, the synthetic phagocytosis-stimulating tetrapeptide (L-threonyl-L-lysyl-L-prolyl-L-arginine), on the nitrous blue tetrazolium (NBT) reduction by human polymorphonuclear leukocytes was investigated. It was found that this substance increases the NBT reduction by approximately as much as endotoxin. Other tetrapeptides do not share this property. When Tuftsin analogs are ...
متن کاملTuftsin deficiency in AIDS.
Tuftsin is an endogenous tetrapeptide that stimulates phagocytosis and is released from the Fc fragment of IgG by a splenic endocarboxypeptidase. Tuftsin activity and splenic function were measured in 21 patients with AIDS, 7 patients with AIDS-related complex (ARC), 22 patients who had undergone splenectomy, and 37 healthy volunteers. There was a significant inverse correlation between tuftsin...
متن کاملInteractions of tuftsin with bovine serum albumin.
Interactions of tuftsin (Thr-Lys-Pro-Arg) with bovine serum albumin (BSA) were analysed by fluorescence spectroscopy and circular dichroism. The data show that tuftsin interacts weakly with BSA, but this interaction is considerably enhanced by introducing an apolar substituent at the C-terminus of the tetrapeptide. It is suggested that strong binding of tuftsin to albumin in blood may enhance i...
متن کاملFunction of Neutrophils in Different Phases of Chronic Myelogenous Leukemia
In chronic myelogenous leukemia (CML), the mature granulocytes originate from a stem cell line harboring an abnormal chromosome, therefore it is possible that metabolic-functional abnormalities occur in the morphologically mature cells. In the present study, the phagocytic activity including intracellular killing, nitro blue tetrazolium (NBT) reduction, and phagocytosis were studied in 37 CML p...
